桿狀病毒蛋白基因工程以及流感仿病毒的研究與應用

本實驗室專注於高表現基因及啟動子的瞭解與調控，同時也將此基礎研究上應用到各種疫苗及擬人化蛋白質的生產等。我們選擇昆蟲桿狀病毒為主要的研究工具之一，因為它並非人類病毒，因此安全而容易操作。由於此病毒在昆蟲細胞製造蛋白質的品質優於細菌、酵母菌，可適用於醫學、生物科技之高標準的需求。並和廠商合作以生產重要疫苗，如禽流感病毒疫苗等。進而也發展出以桿狀病毒早期基因IE2 活化CMV 啟動子，可在哺乳類細胞上研究應用。我們進一步發現IE2 能在細胞核內形成一個個的奈米球狀的結構，可包入肌動蛋白及RNA 聚合酶等轉錄所需的種種蛋白，而形成一個在光學顯微鏡下就能觀察並研究的動態基因表現構造。它是一個新奇的生物微型機器，可為細胞的DNA感受器，並以新穎方式促進基因表現。此系統的發現也使得本實驗室在理論研究及應用上具獨特的競爭優勢。在研究病毒與寄主細胞的互動中，不同於一般相信 miRNA 只影響 mRNA，我們發現miRNA 可控制 DNA 的複製。這個發現將來對miRNA 及DNA 複製的研究會有重要的影響。同時，我們也以創新的方法整區塊分析蛋白質結構，以改造其穩定性，此在酵素活性及疫苗效性的研究及應用上非常重要。本實驗室從創新的基礎研究出發，並注重應用價值，是一個具未來性的實驗室。
